Molecular and serological detection and characterization of Brucella species in Tanzanian livestock

Описание:Brucellosis is a major zoonotic infection in many low- and middle-income countries, yet there is often a lack of information about which Brucella species are responsible for human and livestock disease. Previous studies in Tanzania have reported the presence of Brucella spp. but there is limited data to demonstrate which species are responsible for brucellosis in different livestock species, or concerning transmission patterns among animals. We report serological and molecular data from livestock in districts of northern Tanzania, selected to represent pastoral, agro-pastoral and periurban livestock keeping systems. Data collection at selected households included a survey and the collection of diagnostic samples (blood and vaginal swabs) from livestock (sheep, goats and cattle). Serological testing for smooth Brucella spp. (notably B.abortus and B.melitensis) included competitive enzyme-linked immunoassay (cELISA). Livestock sera were also tested using an indirect ELISA (iELISA) employing a rough strain antigen. Molecular testing of livestock swabs was performed using quantitative PCR (qPCR), with molecular typing of positive samples using a qPCR based single nucleotide polymorphism (SNP) typing method and multi-locus sequence typing (MLST) where possible.
