Bulk segregant analysis (BSA) for the detection of AFLPs linked to the PVX resistance gene in Solanum commersonii

Descripción:The Bulked Segregant Analysis (BSA), in combination with AFLP molecular markers, allowed the identification of DNA polymorphisms in the wild potato Solanum commersonii associated to the resistance gene of the Potato Virus X (PVX). Initially, the F2 individuals were divided in 2 groups, by the means of an ELISA test, one group formed with the resistant individuals and another with the susceptible ones. Later, for the BSA, the mix of DNA from all resistant individuals in one side, and the DNAs from all susceptible ones on the other, were independently analyzed using 64 different primer combinations. By analyzing the bands obtained, 22 different primer combinations yielded AFLPs related to the PVX resistance trait.
