Discovery of a New Specific Bacteriophage for Brucella canis

| Beskrivelse: | In this study, a specific bacteriophage was isolated against Brucella canis, which has become a major problem in dogs in many countries around the world. A lytic phage was isolated from vaginal swabs and litter/manure from a suspected infected dog shelter. Serum samples from dogs living in this shelter were evaluated with home-made ELISA prepared with both rough and smooth Brucella antigen and it was revealed that there were positive and suspicious animals for Brucellosis. Because of serological positivity, vaginal swabs and litter/manure samples were cultured for Brucella spp. isolation. In the culture method, Brucellaspp. could not be isolated. Purification of the phages was performed using the double layer agar method. The propagation of isolated phage yielded high titre of phages (1019 Plaque forming unit, PFU) and the lytic activity of the phage was analysed in different Brucella reference strains. Although the isolated phage showed a lytic effect on B.canis RM6/66 and B.abortus 45/20 strains, it showed no lytic effect on B.abortus (bv1,2,3,4,5,6,9), B.melitensis (bv1,2,3), B.suis (bv1,4,5), B.inopinata, B.ceti and B.abortus s99 strains. Since the phages isolated in the study lysed some rough Brucella strains, their protein structures were comparatively analysed using the SDS-PAGE method with the standard rough R/C phages. As a result of the analysis, it was found that both phages exhibited a common protein profile. Although the protein region close to 70 kDa was prominent in the R/C phage, it was absent from the shelter phages isolated in this study. However, to analyse the phage at the genomic level, the phage DNA was extracted using a commercial phage DNA extraction kit and sent for whole genome sequencing analysis. Bacteriophages are considered promising in biological tools in combating disease agents, including brucellosis. To become more effective, multiple phage cocktails could be developed by targeting multiple hosts. |
