Development of an ex vivo IFN-γ assay for porcine brucellosis diagnosis

| বিবরন: | Porcine brucellosis, caused by Brucella suis biovar 2 (BS2), presents a significant challenge in Europe, particularly due to its presence in wild boar populations which act as reservoirs. While Belgium is officially brucellosis-free in livestock, the disease is endemic in wild suids, posing threat of spillover to domestic pigs. Current serological surveillance in pigs is complicated by false-positive reactions (FPRs) due to cross-reactivity with other bacteria, notably Yersinia enterocolitica O9 (YEO9), highlighting the need for a more robust confir- matory diagnostic tool. This project aimed to develop an ex vivo IFN-γ assay to confirm serological results in porcine brucellosis diagnosis, addressing the absence of injectable brucellin. Additionally, we sought to establish a serum bank from experimentally infected animals to evaluate commercial ELISA kits. We conducted experimental infections on two groups of pigs: one with BS2 and another with YEO9 to induce FPRs. Brucellin was purified from Brucella melitensis B115 and a modified rough strain of Brucella suis. Bacteriological and serological monitoring was performed. For the IFN-γ assay, peripheral blood mononuclear cells (PBMCs) were isolated from infected animals, stimulated with purified antigens, and IFN-γ production was measured by ELISA. Experimental infection with YEO9 successfully reproduced FPRs in standard serological tests and demonstrated with bacterial presence confirmed post-mortem in 90% of the animals. BS2 infection led to seroconversion in all pigs and post-mortem bacterial isolation in 80%. Preliminary IFN-γ assays showed promising results, with detectable production in infected groups. In conclusion, We successfully established experimental infection models for both BS2 and YEO9, effectively mimicking natural infection and FPR scenarios. The preliminary results of the ex vivo IFN- assay are encouraging for a confirmatory diagnostic tool. |
